When connected to the computer you must read from the radio first then you can program it and then send it back to the radio.

Hi Phil,
Are you using a traditional serial port for your programming cable or a USB programming cable? If USB, have you used it with other radios or is this the first time? Many USB cables have a USB-to-serial chip in them that requires that you install an older driver for it to work. Visit the miklor.com website to find out how to get the correct drive and install it.
Also, a common issue for the UV-5R is that the user is fooled into thinking that the cable is fully plugged in when it isn't. Give it an extra wiggle and push to make sure it is all the way in. I'm not kidding, this is a very common problem.
