Larry, It sounds like you have the same windows issue as Bill (in the last post) :- )
As a courtesy to users, Windows 10 updates drivers to the latest version.
Unfortunately, this is not what you want with a Prolific cable.
The drivers you need (3.2.0.0) can be found at http://www.miklor.com/COM/UV_Drivers.php
Also, the driver does not follow the cable. If you change USB slots, you must also load 3.2.0.0 to the new COM port.
That along with the Latest Daily Build of CHIRP and you should be up and running.

CHIRP was working until today's software upgrade. I uninstalled and reinstalled once I could not clear the error.
When attempting to "Upload from radio" the pop-up box appears correctly confirming COM port, vendor and model. After selecting OK,
Then "error #2" pops up "could not open port COM3. The system cannot find the file requested."
Windows 10 - Device Mgr tells me that COM3 is set for the Prolific USB-to-Serial. Under General, "This device cannot start (Code 10). A device which does not exist was specified. Prolific driver is 9/4/2015, version 3.6.81.357
