No, user must be member of dialout group to be able to access serial port devices:
marco@somewhere:~$ ls -l /dev/ttyS0 crw-rw---- 1 root dialout 4, 64 ott 17 08:28 /dev/ttyS0
I strongly suggest you to use root user only for administrative task.
